Génétique et physiopathologie des MNM liées à la matrice extracellulaire et du noyau

Génétique et physiopathologie des Maladies Neuromusculaires liées à la matrice extracellulaire et du noyau

Les thèmes de recherche de l’équipe se concentrent sur 2 groupes de maladies neuromusculaires: les myopathies dues aux anomalies de la Myomatrice (collagène de type VI & autres composants de la matrice extracellulaire) et du Myonucleus (dystrophie musculaire d’Emery-Dreifuss & autres laminopathies du muscle strié dues aux mutations du gène codant pour des lamines de type A ou des gènes codant pour des composants liés à la membrane nucléaire). Ces myopathies partagent certaines caractéristiques cliniques, dont des rétractions importantes, et peuvent faire l’objet d’un diagnostic différentiel.

Ces pathologies sont très hétérogènes, à la fois cliniquement et génétiquement, et aucun traitement n’est encore disponible. Nos activités antérieures ont conduit à l’identification d’anomalies génétiques et au développement d’outils (modèles cellulaires et animaux) afin de décrypter les mécanismes physiopathologiques pour une meilleure compréhension des bases moléculaires et l’identification de cibles thérapeutiques.

De nombreux points restent non résolus: 1) l’absence de diagnostic moléculaire pour un sous-ensemble de patients, 2) l’absence de biomarqueurs pertinents, 3) la fonction précise de certaines protéines impliquées et les mécanismes physiopathologiques sous-jacents… Plusieurs points communs (dysfonctionnement contractile, défauts de la mécanobiologie, fibrose…) ont été et sont toujours abordés de manière transversale par la mise en commun de nos compétences spécifiques (enveloppe nucléaire, nucléoplasme, matrice extracellulaire…).

Thèmes de recherche actuellement développés en parallèle et en synergie

  • Réduire l’errance diagnostique des patients présentant ces MNM par des approches multi-Omics en collaboration avec le Consortium Européen Solve-RD,
  • (Re) Définir le spectre génétique et clinique ainsi que l’histoire naturelle de ces MNM,
  • Développer de nouveaux outils d’aide à la validation de variants génétiques issus du séquençage à haut débit ;
  • Etudier les mécanismes physiopathologiques des mutations géniques qui induisent ces affections des muscles cardiaques et / ou squelettiques avec pour objectif,
  • Identifier et tester des pistes thérapeutiques pour ces affections. Ceci est effectué sur du matériel biologique provenant de patients (ADN, ARN, cellules en culture et biopsies) ainsi que sur les divers modèles animaux que nous avons développés (souris, poisson zèbre).
